Masspropagation of Nephrolepis exaltata by Runner Tips in vitro

Abstract
The effects of inorganic nutrient, growth regulators and light or dark treatments on organogenesis and development of runner tips of Nephrolepis exaltata cv. Bostoniensis and Scotti were investigated for year-round mass propagation.
Addition of sucrose to the medium exerted an important role on frond development from runner tips, and fresh weight and number of propagules were increased at 2¡­3%. The development of fronds was possible under light condition, and that of the roots was feasible under dark.
Multiple shoot formation was obtained on the full strength Murashige-Skoog medium with 200 §·/§¤ NaH©üPO©þ¡¤H©üO, but good root formation and growth was achieved on the half strength MS medium with reduced phosphate concentration.
Kinetin and Zip were more efficient in organogenesis and development than BA treatment. Kinetin was effective in multiple shoot formation, and Zip was promotive in shoot growth and leaf development with effective concn of 1¡­2.0§·/§¤ for Bostoniensis and 0.5§·/§¤ for Scotti.
Combination treatment with NAA or IBA was more effective in organogenesis and development than the single treatment of 2ip or kinetin, with better results obtained by NAA. The mixture of kinetin and 1¡­1.5§·/§¤ NAA and that of Zip and 0.5§·/§¤ NAA promoted fresh weight and the number of propagules.
The successive subculture up to 5¡­6 times reduced organogenesis, indicating the necessity of culturing new runner tips for year-round micropropagation.
